低电压激活(LVA) T型钙通道的特点是瞬态电流和触发神经元放电和振荡行为的低阈值峰值(LTS)。结合T型钙离子通道在树突中的优先定位和特定的“窗口电流”,T型钙离子通道被认为是信号放大和突触整合的关键因素。在新兴药理学工具的辅助下,通道门控和动力学的结构决定因素,以及T型钙通道的新的生理和病理功能正在被揭示。本文综述了T型钙离子通道的结构决定因素、T型钙离子通道在疾病和疾病中的作用、新型通道调节剂的研究进展,以及有助于合理设计药物的构效关系(SAR)研究进展。
