金刚烷基团是许多化合物的结构骨架,它的发现在化学方面推进了一个新领域的出现,该领域研究多种合成的方法,以及有机多面体化合物的理化性质和生物学性质,并实际应用到医药工业。金刚烷的衍生物已被证明是很有效的化合物,从系统性到局部性的治疗有很广的应用范围。本文综述了目前在临床实践中所用的金刚烷衍生物(金刚烷胺、金刚,金刚乙胺,曲金刚胺,阿达帕林,沙格列汀,维格列汀),专注药物的作用机制、药代动力学、药效学和临床试验。文中所提到的基于金刚烷的化合物都已被批准广泛用于一些适应症(抗病毒、降血糖、抗老年痴呆症和帕金森氏病)。对于世界各地众多的患者,每一个化合物都已被证明在适应症的治疗方面是至关重要的。这篇评论还考虑到了副作用的机制,以提供一个完整的视角来看待目前的治疗选择。

摘要: The adamantane moiety is the structural backbone of numerous compounds and its discovery launched a new field of chemistry studying the approaches to the synthesis as well as the physicochemical and biological properties of organic polyhedral compounds with practical application in the pharmaceutical industry. Adamantane derivatives have proven to be very potent compounds in a wide range of applications from systemic to topical therapy. This review summarizes the currently available adamantane derivatives in clinical practice (amantadine, memantine, rimantadine, tromantadine, adapalene, saxagliptin, vildagliptin), focusing on mechanisms of action, pharmacokinetics, pharmacodynamics and clinical trials. The adamantane-based compounds presented in this manuscript have been approved for a wide spectrum of indications (antivirals, antidiabetics and against Alzheimer’s and Parkinson’s disease). Each of the compounds proved to be of vital importance in their therapeutic indication for numerous patients worldwide. This review also considers the mechanisms of side effects to deliver a complete perspective on current treatment options.
