摘要
Hedgehog(Hh)信号通路是一种发育的信号通路,是许多脊椎动物系统正常胚胎发育必要的。Hh信号通路的失调是人类多种肿瘤的发生发展过程中的一个因素。因此,作为潜在的抗癌药物的小分子Hh信号抑制剂的发展已成为学术界和产业界在过去十年的主要研究方向。通过这些努力,Hh信号通路多靶点的合成小分子已被鉴定并进入临床前或临床开发。本文的目标是提供对几种合成小分子Hh通路抑制剂现状的信息更新并探索几种最近披露的抑制支架的潜力。

摘要: The hedgehog (Hh) pathway is a developmental signaling pathway that is essential to the proper embryonic development of many vertebrate systems. Dysregulation of Hh signaling has been implicated as a causative factor in the development and progression of several forms of human cancer. As such, the development of small molecule inhibitors of Hh signaling as potential anti-cancer chemotherapeutics has been a major area of research interest in both academics and industry over the past ten years. Through these efforts, synthetic small molecules that target multiple components of the Hh pathway have been identified and advanced to preclinical or clinical development. The goal of this review is to provide an update on the current status of several synthetic small molecule Hh pathway inhibitors and explore the potential of several recently disclosed inhibitory scaffolds.
